The longer run, and when it is the right one
Ostuni is the white town on the hill above the olive plain, and it sits closer to Brindisi than to Bari. From Bari Airport the drive is roughly an hour to an hour and a quarter — an estimate from the road distance rather than an operator-published time. For comparison, Brindisi Airport to Ostuni is 37.7 km and about 31 minutes.
That is worth knowing at the point you book flights, not afterwards. If Bari gives you a better fare or a better time, take it: an hour in a comfortable car with your luggage in the boot is not a hardship, and the same companies cover both airports. If the flights are equal, Brindisi wins on this route.
Price, labelled honestly
None of the verified Puglia chauffeur companies publish per-route fares — they quote from a form, which means the only public numbers on this coast come from the official airport taxi tariff and the aggregators. Two anchors to think with: private transfers on the shorter Brindisi to Ostuni run are listed at roughly 123 to 149 EUR, and the Bari to Polignano route carries a fixed official airport taxi fare of 80 EUR with aggregator listings around 104 to 110 EUR. Bari to Ostuni is a longer drive than either. We come back with a real quote rather than an extrapolation.

Vehicles, luggage and children
The fleets run Mercedes E-class sedans for up to 3 passengers, V-class and Vito vans for 7 to 8, and Sprinters for bigger groups, with wifi in the vans and English-speaking drivers. Send the luggage count as well as the passenger count. Child seats are supplied free by at least one Bari company and on request by the others, and Italian law requires child restraints in cars — ages and weights with the booking, please, because seats are fitted before you land.
Finding a masseria in the dark
This is the real argument for a pre-booked local driver on this route. Ostuni’s countryside accommodation sits down unnamed lanes between olive groves, and the last kilometre defeats satellite navigation regularly. Send the property’s own arrival notes with your request and they go to the driver in advance.
The verified companies cover Bari airport, the cruise terminal and the station through to Polignano, Monopoli, Ostuni, Alberobello, Matera and Brindisi airport — so an arrival at one airport and a departure from the other is one booking.
